I read that the pik3ca mutation, which gdc0077 targets, is most prevalent in Er or Pr positive subgroup of breast cancer, and makes up 34.5% of that group. Others groups, like TN have it as well, but at a lower frequency (8.3% TN, 22-31% for HER2+).

Hubby- PI3K mutations are the most common way cancers grow around estrogen deprivation. If you can block that pathway, then the cells can go back to being estrogen-dependent again. But, it's been hard to get a good drug in this space. I've read that it may be best to use this drug secondline or later, to increase the fraction of cancer cells that carry the mutation. Am following Babs with interest, and so glad it seems to be working!

Got my scan results and wanted to share them with you. My liver mets decreased as follows in 2 months time :
Segment 5 went from 2.1x2.1 to 1.4x1.3
Segment 6 went from 2.5x2.1 to 1.6x1.3
Segment 2 went from 2.8x2.6 to 2.4x2.3

Back today for the study
Weight oxygen glucose blood pressure 2 vials of blood
Met with study nurse. Asked her how others in the study are doing. As of now everyone is seeing results. Met with dr. All is good
Back in 2 weeks

I’m so happy for you! I had to go off of it, it raised my glucose too high too fast. I was only able to get 5 doses in. Plus it caused many other side effects for me. I’m done doing clinical trials for a bit. My oncologist said my body needs a rest.

I had severe fatigue, ringing in my ears, my sodium levels dropped way too low, my potassium also dropped, I felt awful, it took over a week to start to feel normal again.

I'm also fatigued but have been for so long due to all the chemos. Plus working FT and going out so much at night. Never thought it was from the GDC0077

I also have the normal fatigue from chemo and working full time. I work in a school with special needs children. The fatigue I had with the GDC was extreme, I literally had to keep laying down every half hour or so or I’d feel like I was going to pass out. I would actually get cold, clammy and dizzy.

Today was my study day. Took weight, blood pressure, temperature EKG oxygen Then they took my blood. Since I've been real tired I asked them to have a nurse who always can get my blood on the first shot. Well they did and she did! Afterwards I went down to see my NP and there was the nurse from clinic again. She was not given the full 10 vials before taking my blood. So she had to take more blood. Ouch. Yes I'm black and blue
All seemed good until I got my TMs today The ca15-3 and Cea both went up. I have a call Into my MO. Will keep you posted

How much are CEA and CA 15-3 up? Is this the first jump or is it a trend?
I hate getting blood taken. Sorry for the two pokes.

z Cea went from 46 to 67
Ca 15-3 went from 354 to 391
The first 28 days they went down by 1/3. Next 28 days they went down by about 12%. Mo wants to wait for scan on 21 I'm actually off the GDC0077 due to starting rads on Monday. Ugh!
I hate this roller coaster ride we're all on

Not a huge change. Hugs and strength as you deal with rads and wait for scan. It seems wise to wait but the waiting is hard. I hate the roller coaster.

Went to MSKCC on Thursday. Since I'm off the GDC0077 while doing rads all that was done was weight, temp, oxygen levels and 2 vials of blood. My Liver levels have shot up. Finished rads yesterday so go back on the GDC0077 Monday morning. Scheduled scan on Tuesday. Fingers crossed but not expecting good results!
