Palbociclib - side effects???

Options

Can any of you taking this drug already shed some light on any of the side effects ? I am just about to go into a clinical trial for stage 2/3 ER/PR positive BC combining hormone therapy and Palbociclib for 2 years. I have all the literature on potential side effects but first hand advice would be appreciated. I will be combining it with Tamoxifen for a few months before switching over to an aromatase inhibitor.     I can't help you with that but it sounds like an interesting approach. I appreciate that you're doing this and hope it goes well for you. I'd love to read updates from time to time.

    I began the Feasibility trial for Palbociclib and made it through 6 of the 24 cycles, with 2 dosage reductions. As we all know, everyone has different SE's from any treatment. Hopefully, you will have minimal side effects! Palbo was rough for me. I had Neutropenia, intense fatigue, and hair loss. I believe that it triggered my migraines, which had been under control. (Trial nurse said others experienced this as well.) Weirdly, I developed internal hemorrhoids which disappeared when I came off Palbo (not sure if related). I burped a lot. There is a Stage IV thread regarding Palbo. Some of the stage IV participants are/were receiving a placebo (including the original poster). Some had few, if any, side effects. I was discouraged not to make it the full 2 years. I wish you success. If you have specific questions, please PM me. (From the stage IV thread I learned the importance of taking it with food.)

    Thanks, Daisy... I am worried about the fatigue. The trial nurse said not everyone gets it but,as I have a very active job I can't afford to let that be an issue. I have the winters off so, I thought trying it now would be the best . I see you are from MA ... Me too.

    Lenn, I think if you do not get the Neutropenia, you probably won't get the fatigue? I had 3 colds during my short time on Palbo! (hadn't had a cold in years) Luckily, I don't really have any side effects with Tamoxifen! I am probably going to have an oopherectomy and then try an AI as well. Hoping for no serious SE'S then... Hope you are prepared for some snow!

    Same here Daisy. Really no problems with tamoxifen. That's why they want me on the Palbociclib before I switch so I can tell what is a side effect from the aromatase inhibitor and what is from the Palbociclib. I am pretty healthy right now...like you no colds, etc...

    Bring on the snow. I am a winter lover ... Skier, skater, etc just don't like to shovel
