Hi Folks,

I'm presently participating in Phase 2 of the above trial @ MSK and am interested in hearing from others who are participating in this trial.

My background: 2003 at age 44 dx with bilateral breast ca-different cancers in each breast. 100% er/pr+ her2-. 3 rounds of ACT, then d/c'd due to ileus. Tried tamoxifen but it caused cognitive issues, so no maintenance meds. In 2013, experienced swelling of left arm which lead to discovering a mass in my left axilla which lead to a stage iv dx of bone mets. Successfully treated with letrozole for three years until wonky bldwk and a bone marrow bx confirmed infiltration. Was successful on ibrance/faslodex combo for a year (my body responded very well to this combo and my marrow was chugging along. My MO stated my numbers were "remarkable"). Liver lesions in November 2017 scans. PK1 mutation identified and began this trial the end of January.

I am dealing with the hyperglycemia SEs,but with the addition of oral meds and a decrease in the trial med, I feel we have hit the sweet spot and my glucose is under control with many diet changes. I miss my Pepsi. Yes, I drank regular Pepsi every day, all day for close to 40 yearrs!

Tumors markers: CEA was 6.2 prior to starting trial med and now is 5.6. CA 15-3 was 114 and now is 49. All other chemistry results are in normal range. First CT scan is 3/20. I'm holding on to since I have great bldwk results, I'll have a good scan result.

Please share your experiences/results so I can be educated on how others are doing!

    I failed the trial but that happened really while I was off treatment due to rads. I hope it works for a long time for you!

    How/why did you "fail" Babs? Was it the because of blood work and/or CT results? Or because of the inconsistency of taking the trial meds due to your radiation schedule? Or did you have to choose either the trial or radiation? Thanks!

    Both blood tests and scans were why I failed. However I really think that being off the meds during rads played into my progression and increased TMs. But the u increases were high so couldn’t stay in trial and was also worried about the increases
