Anyone doing T-DM1 Clinical Trial in Adjuvant setting?
I've just signed on to join a small clinical trial on T-DM1, which is a combination of Herceptin and another chemo drug to replace the standard Herceptin/Taxol regimen. It's been used with Stage IV metastatic breast cancers for a while (plenty of good response to it over on the Stage IV boards), but is just now being studied in candidates with earlier or locally advanced breast cancers as an adjuvant therapy. It is a worldwide study: not many slots in the U.S., but I noticed New York and San Antonio, as well as Boston, on the list. Just wondering if any one else has heard of it, or been given the opportunity to participate.

This is so interesting. I know I even asked my onc about T-DM1 and he advised me that I didn't qualify because of my stage. Which of course, is a nice thing to hear, but the results have been so good that I always thought it would be nice if it were offered to other stages. Can you tell me where to find out more information about this small trial that involves ladies with stages other than 4? As you can see, I am similar to you.

And yes - they want early stage or locally advanced early stage HER+2 for this run. There aren't many slots apparently, but they are recruiting. You might check the link at Genentech for more info?

Yes, girlstrong. I am supposed to start the Katherine trial at my next treatment on 4/30. I am getting them in Lynchburg, VA at the Alan B. Pearson cancer center. There are side effects that I'm nervous about getting, but you can always withdraw your consent if you get too sick. Plus, who know if you'll even be getting the new drug, right?
By the way, this is my first post. Ive searched the world over for someone else doing this and you are the only post I can find.

hi Celesteva, I think that we may be a rare group....I had heard that there are only 100 women worldwide in the study so far. Not sure if that's true??? I met with a new onc yesterday and if I am accepted into the trial I will need to transition my care to Karmanos cancer center in Detroit, Mi since they are an approved site for the trial. So I signed all the consents and had blood drawn just yesterday. Had a MUGA scan done earlier in the week etc. the only reason I may not be accepted into the trial is if I go past the 12 weeks post surgery cut off point. Right now I'm at week 10; the new onc says that I still have a good chance since "they" really want and need subjects for this study and so it's not uncommon for studies to make exceptions. Jut finished up TCHP (6 rounds) and have completed 2 rounds of herceptin. I am 3 wks post BMX. I am meeting trial coordinator tomorrow to sign up for KATHERINE. Won't know what arm I get into but MO wants me to get into the TDM1 side. Is anyone still in or has compledted the trial? Any advice on SEs or have they been minimal...Thanks.

Hi larod825, I am in the KATHERINE trial and down to my last three treatments (of 14). I was randomized to Kadcyla. For me, infusions 2-4 were pretty rough, with flu-like symptoms, elevated liver numbers (and right-side pains) and a decreased EF. But I bounced back around cycle 5 and I've barely had side effects since. Fatigue is the only complaint and I've been in treatment since Jan. 2014 so that's to be expected!
I'm glad I had the opportunity to do a trial. Even if the Kadcyla turns out to do nothing for my prognosis, I'm happy I was able to contribute to research and help other women.

I am going to talk to my onco tomorrow about this trial. I finished TCHP in February and tomorrow will be my 4th herceptin alone (10 if you include it during chemo) so I'm nervous about changing what I am doing now. The herceptin gives me hip and knee joint pain pretty bad but I am pretty active and being only 33, I bounce back pretty quickly when I feel bad. Does anyone know where I can get more information on tdm1

so this is something done after you finish chemo I have done 2 out of 6 tchp and then I will of course be on herceptin for 1 year this trial has herceptin and another drug with it? I wonder if by the time I finish chemo it will be too late to join the trial last one is scheduled in july

We were aware of the trial and my wife's medical oncologists at the James in Columbus recommended it. She called T-DM1 a "cancer bomb"! They are so optimistic about the trial that three out of four women in the trial will receive the T-DM1 treatment as opposed to the 1 of 4 who will receive the "standard of care" treatment of Taxol/Herceptin.
Unfortunately my wife was disqualified when an initial IHC test post surgery showed her as negative for HER2. Follow up FISH testing still indicates HER2 positive because of copy number.

I am in the KATHERINE trial. I have my 6th (out of 14) treatments tomorrow. I was randomized to Kadcyla. I was stage 2a and had a BMX after 6 cycles of TCHP

I just signed up for it, they are doing the trial at University of Pittsburgh Medical Center. I refuse to read anything online anymore, as it only created anxiety... but did read some nasty side effects of TDM1 and it scares me. I just had my 10th round of herceptin and do okay on that. It's tough making a decision like this! I may back out but not sure what to do.

Iwill be starting a clinical trial on July 1st, after my 4 doses of AC. I will be getting Kadcyla and perjeta every three weeks for one year. I was selected into the trial and randomly selected for Kadcyla/Perjeta vs Taxol/Hercepton/Perjeta. This trial is in phase three.
